1. What is Vonocab® Tablet?
Vonocab® is a potassium competitive acid blocker (P-CAB).
2. How Vonocab® Tablet works in our body?
Vonocab® is a potassium-competitive acid blocker with a unique mechanism of action.
Vonocab® is a potassium-competitive acid blocker. Potassium (K+) plays an important role in the activation of H+, K+ ATPase (proton pump). When potassium (K+) enters the parietal cell through the potassium channel, proton (H+) goes out of the parietal cell to the gastric lumen in exchange for potassium (K+) and combines with chloride (Cl–) to form hydrochloric acid. Vonocab® blocks the potassium (K+) ion channels in H+, K+ ATPase (proton pumps). Thus, potassium (K+) cannot enter the parietal cell, and proton (H+) cannot go out to form hydrochloric acid. Thus, by blocking the potassium binding site of H+, K+ ATPase (proton pumps) Vonocab® inhibits acid secretion.
3. What is the indication of Vonocab® Tablet?
Vonoprazan is indicated for:
- Gastric & duodenal ulcer
- Reflux esophagitis (GERD)
- Prevention of recurrence of gastric or duodenal ulcer during low-dose aspirin or non-steroidal anti-inflammatory drug (NSAID) administration
- Adjunct to Helicobacter pylori eradication
4. What is the dosage & administration of Vonocab® Tablet?
Gastric and duodenal ulcer: The usual adult dosage for oral use is 20mg of Vonoprazan administered orally once daily 8 weeks for gastric ulcer and 6-week treatment for duodenal ulcer.
Reflux esophagitis: The usual adult dose for oral use is 20 mg of Vonoprazan administered once daily for a total of 4 weeks of treatment. Treatment can be extended to 8 weeks if required. For the maintenance therapy of reflux esophagitis, the dose for oral use is 10 mg of Vonoprazan once daily. However, when the efficacy is inadequate, the dosage may be increased up to 20 mg of Vonoprazan once daily.
Prevention of recurrence of gastric or duodenal ulcer during low dose aspirin & non-steroidal anti-inflammatory drug (NSAID) administration: The usual adult dosage is one tablet of 10 mg of Vonoprazan administered orally once daily.
Adjunct to Helicobacter pylori eradication: For adults, the following three-drug regimen should be administered orally at the same time twice daily for seven days:
20 mg of Vonoprazan,
1000 mg of Amoxicillin hydrate
500 mg of Clarithromycin
5. What are the contraindications of Vonocab® Tablet?
Vonoprazan is contraindicated in patients with hypersensitivity to Vonoprazan or to any excipient of the product.
6. What are the adverse effects of Vonocab® Tablet?
Following adverse reactions have been reported with the use of Vonoprazan: Diarrhea, constipation, Nausea, Bloating, Jaundice, Anaphylactic Shock, Liver function test abnormal, Adema, AST increased, ALT increased, LDH increased, eosinophilia.
7. What would be the storage condition of Vonocab® Tablet?
Do not store above 30°C. Do not refrigerate or freeze. Keep in a dry place. Protect from light & keep out of the reach of children.
